en سلامت جانبازان يا معلولان Veterans or Handicapped Health پژوهشی اصيل Original Research <span style="font-size:12pt"><span style="line-height:normal"><span style="font-family:Calibri,sans-serif"><b><span new="" roman="" style="font-family:" times="">Background:</span></b><span new="" roman="" style="font-family:" times=""> <i>Helicobacter pylori</i> remains one of the most prevalent human bacterial pathogens, colonizing approximately 30&ndash;70% of the global population and representing a major cause of chronic gastritis, peptic ulcer disease, and gastric adenocarcinoma. This study evaluated the in vitro antibacterial activity of date seed extract against a multidrug-resistant clinical isolate of <i>H. pylor</i>.</span></span></span></span><br> <span style="font-size:12pt"><span style="line-height:normal"><span style="font-family:Calibri,sans-serif"><b><span new="" roman="" style="font-family:" times="">Methods:</span></b><span new="" roman="" style="font-family:" times=""> A multidrug-resistant clinical isolate of <i>H. pylori</i> was recovered from a gastric biopsy specimen obtained from a dyspeptic patient and identified by conventional microbiological tests and PCR amplification of the <i>cagA</i> virulence gene. Ethanolic date seed extract was prepared under standardized laboratory conditions. The phytochemical profile of the extract w:as char:acterized by GC&ndash;MS analysis. Antibacterial activity was evaluated using the agar well diffusion assay, while the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined using the resazurin microdilution assay and subsequent subculture, respectively. All experiments were performed in triplicate and expressed as mean &plusmn; standard deviation.</span></span></span></span><br> <span style="font-size:12pt"><span style="line-height:normal"><span style="font-family:Calibri,sans-serif"><b><span new="" roman="" style="font-family:" times="">Results:</span></b><span new="" roman="" style="font-family:" times=""> Phytochemical profiling identified eighteen major bioactive compounds in the ethanolic extract of Phoenix dactylifera seeds, mainly proanthocyanidins, flavonoid glycosides, catechins, and hydroxycinnamic acid derivatives. Proanthocyanidin tetramer was the predominant constituent. The extract exhibited pronounced antibacterial activity against the multidrug-resistant <i>H. pylori</i> isolate, producing an inhibition zone of 32.0 &plusmn; 0.2 mm. The MIC and MBC values were 15.6 &plusmn; 0.5 &mu;g/mL and 31.2 &plusmn; 0.3 &mu;g/mL, respectively.
